报告摘要:
Genetic variants underlying both complex and Mendelian diseases may be affected by natural selection and conversely, regions of the human genome under natural selection are likely to harbor functional loci that may influence disease susceptibility. Evolutionary forces are likely to have operated on common diseases such as coronary heart disease and inferences regarding natural selection may therefore help identify disease susceptibility loci in humans and facilitate disease-mapping studies. I present an evolutionary perspective of coronary heart disease, a major cause of morbidity and mortality in the world and discuss the current hypotheses and knowledge regarding the evolutionary genetics of several risk factors for coronary heart disease including hypertension, dyslipidemia, and the metabolic syndrome. Several candidate genes in pathways of blood pressure, glucose and lipid metabolism, blood clotting, and inflammation that may be under natural selection are enumerated. An evolutionary perspective might help explain why contemporary humans are at high risk for coronary heart disease and also better understand inter-individual variation in disease susceptibility.
